5-Amino-1MQ
5-Amino-1-Methylquinolinium
The NNMT inhibitor that targets stubborn fat at the cellular level
5-Amino-1MQ is a small molecule that inhibits NNMT, an enzyme linked to fat cell expansion and metabolic dysfunction. It represents a novel approach to fat loss that works at the epigenetic level.

Overview
5-Amino-1MQ is not technically a peptide - it's a small molecule NNMT (nicotinamide N-methyltransferase) inhibitor. However, it is widely sold and discussed in the peptide community. NNMT is an enzyme that is overexpressed in fat tissue, particularly visceral and 'stubborn' fat. High NNMT activity is associated with obesity, metabolic syndrome, and resistance to fat loss. By inhibiting NNMT, 5-Amino-1MQ reduces fat cell size, promotes fat oxidation, and may improve metabolic function. It was developed by researchers at the University of Texas and has shown promising results in animal models.
⚙️How it works
NNMT normally methylates nicotinamide (a form of vitamin B3), consuming SAM (S-adenosylmethionine) in the process. High NNMT activity in fat cells depletes SAM, which reduces the cell's ability to perform methylation-dependent processes including lipolysis regulation. By inhibiting NNMT, 5-Amino-1MQ restores SAM levels in fat cells, promoting fat breakdown and reducing fat storage. It also appears to increase NAD+ availability (since less nicotinamide is being methylated away), creating a link to the NAD+/sirtuin longevity pathway.

A novel mechanism
Most fat loss compounds work through appetite suppression (GLP-1 agonists), metabolic rate increase (thyroid hormones), or fat oxidation (GH, AOD-9604). 5-Amino-1MQ takes a different approach - it targets the epigenetic machinery within fat cells themselves. By inhibiting NNMT, it changes how fat cells regulate their own metabolism. This makes it potentially useful for 'stubborn' fat deposits that resist traditional approaches. However, it's important to note that all data comes from animal studies - no human clinical trials have been published.
